In 2015, Castro was traded to the New York Yankees, where he played as a second baseman. He played with the team until 2019, where he had several ups and downs in his career. He hit .307 in his first season, but his performance dwindled over time. He spent the next few seasons struggling with injuries and inconsistency, hitting just .270 in 2018. In 2019, after returning from an injury, Castro finished the season with an impressive .302 batting average.

In 2020, Castro signed with the Washington Nationals and played as their everyday second baseman. With Washington, Castro found his groove again, hitting .283 with 2 home runs and 38 RBIs in the shortened 60-game season. Despite missing the last month of the 2020 season due to a wrist injury, his performance was enough to earn him a two-year contract extension with the Nationals.
