Williams was selected by the Carolina Panthers in the first round of the 2006 NFL Draft. He played for the Panthers for nine seasons, becoming the franchise's all-time leading rusher with 6,846 yards and 46 rushing touchdowns. Williams was also named to the Pro Bowl in 2009 and was a first-team All-Pro selection in 2008. In 2015, he signed with the Pittsburgh Steelers, where he played for two seasons before retiring in 2017.

Throughout his NFL career, Williams was known for his versatility and ability to run both inside and outside the tackles. He was also a threat as a receiver out of the backfield, finishing his career with 2,931 receiving yards and 13 receiving touchdowns. Williams was also an advocate for breast cancer awareness, having lost his mother to the disease in 2006. He started a foundation in her memory called the DeAngelo Williams Foundation, which provided free mammograms for women in underserved communities.
