Gain played 13 seasons in the NFL, all with the Browns, from 1952 to 1964. During his career, he was a six-time Pro Bowl selection, a two-time NFL champion, and won the Bert Bell Award as the NFL's Player of the Year in 1963. Gain was known for his versatility on the field, playing both offensive and defensive positions, and was dubbed "Mr. Versatility" by his teammates.

After his playing career, Gain became a coach and worked with several NFL teams, including the Buffalo Bills, Oakland Raiders, Los Angeles Rams, and Miami Dolphins. He was inducted into the Cleveland Browns Ring of Honor in 2010, and the University of Kentucky Athletics Hall of Fame in 2019, cementing his legacy as one of the greatest players in NFL history.
