After graduating from high school, Peters chose to attend the University of Washington, where he continued to play football. During his college career, Peters was named a first-team All-American and first-team All-Pac-12 selection. In his junior year, he led the Pac-12 in interceptions with five and was named the Pac-12 Defensive Player of the Year. Peters decided to forego his senior year and declared himself eligible for the NFL draft.

In 2015, Marcus Peters was drafted by the Kansas City Chiefs in the first round of the NFL draft. He had an impressive rookie season, recording a league-leading eight interceptions and earning a spot on the Pro Bowl team. Peters continued to play for the Chiefs for three seasons before being traded to the Los Angeles Rams in 2018. In his first season with the Rams, he started all 16 games and recorded three interceptions, one forced fumble, and two fumble recoveries.
