Darby played college football at Florida State, where he won a national championship in his freshman year. During his tenure with the Seminoles, he emerged as one of the top cornerbacks in the country. As a result, he was a finalist for the Jim Thorpe Award, which is given to the nation's top defensive back. Darby left college after his junior year and declared for the NFL draft in 2015.

Ronald Darby was picked by the Buffalo Bills in the second round (50th overall) of the 2015 NFL Draft. In his first season with the Bills, he started 15 games and recorded two interceptions, 68 tackles, and 21 pass deflections. He was named to the NFL All-Rookie Team that season. Darby played for the Bills for three seasons before being traded to the Philadelphia Eagles in August 2017. He won a Super Bowl championship with the Eagles in the 2017-2018 season. In March 2020, he signed with the Washington Redskins. Throughout his NFL career, Darby has established himself as a reliable and productive cornerback.
