He started his football career at Bingham High School in South Jordan, Utah, where he played as a defensive tackle. He continued his football career at the University of Utah, where he played for four years and was an integral part of the team's defense. In 2011, he was named First-Team All-Mountain West Conference, and in 2012, he was named First-Team All-Pac-12.

He also helped the team reach Super Bowl 50 in 2016, where they lost to the Denver Broncos.
In 2018, Lotulelei signed a five-year contract worth $50 million with the Buffalo Bills. He has continued to be a valuable asset to the team, starting in every game since his arrival.
