In 1959, Grissom was selected as one of the Mercury Seven astronauts who would be the first Americans to go into space. He piloted the second manned mission, Mercury-Redstone 4, on July 21, 1961, which made him the second American in space, after Alan Shepard. The mission lasted about 15 minutes, and Grissom became the first person to experience weightlessness twice. He was also the first astronaut to manually control his spacecraft in space. Later, Grissom also flew in the Gemini 3 mission with John Young on March 23, 1965, which was the first manned Gemini mission.

On January 27, 1967, during a preflight test for the Apollo 1 mission, a fire broke out in the spacecraft that killed Grissom and his fellow crew members Ed White and Roger Chaffee. Grissom had been selected to be the commander of Apollo 1 and was known for his meticulousness and attention to detail. After his death, Grissom was honored by NASA, and awards such as the Congressional Space Medal of Honor and the Grissom Air Force Base were named in his memory.
