Mathews was drafted in the first round of the 2010 NFL Draft by the San Diego Chargers. He played for the Chargers for five seasons and was a valuable member of the team during his tenure. Mathews ran for over 1,000 yards in both the 2011 and 2013 seasons and achieved his career-best 1,255 rushing yards in 2011. Mathews had a reputation as a physical runner with impressive speed and agility, making him an offensive weapon for the Chargers. He was also a reliable receiver and blocker, making him a crucial part of the team's offensive strategy.

In 2015, Mathews signed with the Philadelphia Eagles after leaving the Chargers as a free agent.
